Advant 8mg Tablet

Description

Advant 8mg Tablet (Candesartan cilexetil), a prodrug, is hydrolyzed to candesartan
during absorption from the gastrointestinal tract. Candesartan is a selective
AT1 subtype angiotensin II receptor antagonist.

Candesartan cilexetil, a nonpeptide, is chemically described as (±)-1-
Hydroxyethyl 2-ethoxy-1-[p-(o-1H-tetrazol-5-ylphenyl)benzyl]-7-
benzimidazolecarboxylate, cyclohexyl carbonate (ester). Its molecular
formula is C33H34N6O6, and the structural formula is:

Angiotensin II is formed from angiotensin I in a reaction catalyzed by
angiotensin-converting enzyme (ACE, kininase II). Angiotensin II is the
principal pressor agent of the renin-angiotensin system, with effects that
include vasoconstriction, stimulation of synthesis and release of aldosterone,
cardiac stimulation, and renal reabsorption of sodium. Candesartan blocks
the vasoconstrictor and aldosterone-secreting effects of angiotensin II by
selectively blocking the binding of angiotensin II to the AT1 receptor in many
tissues, such as vascular smooth muscle and the adrenal gland. Its action
is, therefore, independent of the pathways for angiotensin II synthesis.
Candesartan cilexetil is rapidly converted to the active drug, candesartan,
by ester hydrolysis during absorption from the gastrointestinal tract.
Candesartan is an angiotensin II receptor antagonist, selective for AT1
receptors, with tight binding to and slow dissociation from the receptor. It
has no agonist activity. ACE inhibitors also inhibit the degradation of bradykinin, a reaction also
catalyzed by ACE.

Uses of Advant

This medication is used in the treatment of hypertension (increased blood pressure).

Patients using this medication may at times experience dizziness, headache, and back pain. If you experience any of these symptoms for a prolonged period of time, please consult your doctor.

When not to use Advant

This medication is not recommended in patients with severe hepatic (liver) impairment and cholestasis (flow of bile is reduced due to obstruction).
